Distribution of Ca*+ Channels on Frog Motor Nerve Terminals Revealed by Fluorescent o-Conotoxin

چکیده
Tetramethylrhodamine-conjugated w-conotoxin was used as a fluorescent stain (Jones et al., 1989) to determine the spatial distribution of voltage-gated Ca2+ channels along frog motor nerve terminals. Like native w-conotoxin, the fluorescent toxin blocked neuromuscular transmission irreversibly. The fluorescent staining was confined to the neuromuscular junction and consisted of a series of narrow bands (in face views) or dots (in side views) approximately 1 Am apart. This characteristic staining pattern was prevented by pretreatment with w-conotoxin and by prior denervation for 5-7 d. Combined fluorescence and phase-contrast optics indicated that the stain was on the synaptic rather than the nonsynaptic side of the nerve terminal. The bands and dots of stain proved to be in spatial register with the postsynaptic junctional folds, as revealed by combined staining of ACh receptors. It is concluded that the voltage-gated Ca*+ channels on frog motor nerve terminals are concentrated at active zones. The findings are consistent with the suggestion (Heuser et al., 1974; Pumplin et al., 1981) that the large intramembranous particles seen at freeze-fractured active zones are voltage-gated Ca2+ channels.

Spatial Distribution of Calcium Entry Evoked by Single Action Potentials within the Presynaptic Active Zone.
The nature of presynaptic calcium (Ca(2+)) signals that initiate neurotransmitter release makes these signals difficult to study, in part because of the small size of specialized active zones within most nerve terminals. Using the frog motor nerve terminal, which contains especially large active zones, we show that increases in intracellular Ca(2+) concentration within 1 msec of action potentia...

The presence of L-type Ca2+ channels at the frog neuromuscular junction (nmj) was studied by monitoring changes in intracellular Ca2+ evoked in presynaptic terminals and perisynaptic Schwann cells (PSCs) and by studying the distribution of Ca2+ channels using a monoclonal antibody directed against the alpha 2/delta subunit of L channels. L-type Ca2+ channel agonist and antagonist had no effect ...
متن کاملCalcium-dependent depolarizations originating in lizard motor nerve terminals.
Action potentials and afterpotentials were recorded via a microelectrode inserted into motor axons innervating the lizard ceratomandibularis muscle. Intra-axonal injection of Lucifer yellow dye indicated that these axons innervate multiple (at least 6-25) motor terminals. In 10 mM tetraethylammonium (TEA), the action potential was followed by a sequence of afterpotentials, whose amplitude and d...

The transmitter release and synaptic vesicle exo- and endocytosis induced by constant current depolarization of nerve terminals were studied by microelectode extracellular recording of miniature endplate currents and fluorescent microscopy (FM 1-43 styryl dye). Depolarization of the plasma membrane of nerve terminals in the control specimen was shown to significantly increase the MEPC frequency...
متن کاملActive calcium responses recorded optically from nerve terminals of the frog neurohypophysis
Voltage-sensitive dyes were used to record by optical means membrane potential changes from nerve terminals in the isolated frog neurohypophysis. Following the block of voltage-sensitive Na+ channels by tetrodotoxin (TTX) and K+ channels by tetraethylammonium (TEA), direct electric field stimulation of the nerve terminals still evoked large active responses. These responses were reversibly bloc...
